Description: | Calix[4]arene methylene bisphosphonic acid C-192 and its sodium salt C-145 were shown to be efficient inhibitors of fibrin polymerization in vitro (ІС50 was 0,510-6 М and 410-6 М respectably). After 4 hours after intravenous injection of calix[4]arene C-145 to rabbit the thrombin time and activated partial thromboplastin time were prolonged in 2 and 1,5 times respectively. Other parameters of coagulation and fibrinolysis remained constant. Calix[4]arene C-145 also strongly affected platelet aggregation induced by ADP and provoked thrombocytopenia. So we assumed that effects of calix[4]arene C-145 on haemostasis in vivo consists in selectively inhibition of fibrin polymerization and formation of three-dimensional fibrin network that is the core of thrombus. |
